I would say leave the PC as it is (don't shut it off), remove it from the network, and have someone come down to take a look at it, download any antivirus updates, scan the system and backup any data that is needed incase the virus corrupts your hard drive on a restart,etc...

Update and run their anti virus program. Also have them run a couple of spyware scans since some spyware may make some users believe they have a virus. Have them run a online scan which will eliminate if there anti-virus is working.
Superantispyware and spybot run in safe mode would be a good start I feel.
If someone called me with that exact question, I would answer:

Before suggesting anything I would ask why they think they have a virus, for one. It's entirely plausible that they're mistaken. Until there's some kind of rationale, a suggestion is preumptuous.
